You can ask Who is Bart �s sister? if you don �t know who his sister is. I think both the answers you �re suggesting for this question are correct. (Bart �s sister is Lisa/ Lisa is Bart �s sister)
BUT you can also ask Is Lisa Bart �s sister? if you think that Lisa might be Bart �s sister, but you �re not sure. And Yes, she is is the correct answer in that case.
