As a native speaker, I�d say the absolutely correct answer would be "In what is olive oil rich?" However I have never heard anyone actually SAY this..............it would be correct for exam purposes, but in terms of speech we would say "What is olive oil rich in?"

IMHNSO your suggestion is perfectly fine for conversation but if it is to be in a written format I would be inclined to rephrase it as "Olive oil is rich in which vitamins?"
