Depending on context:
 
I�m sure/confident/certain/positive about that
 
Sure, I can do that - Be rest assured I can do that/naturally I can do that
 
Can you do that? - Sure/most certainly/certainly/undoubtedly/without a doubt
 
IMHO using  �of course� is often considered impolite. I once came across a good article on it but now I can �t find it
